Hierbei handelt es sich um Gene, die bereits mit psychiatrischen Erkrankungen assoziiert worden sind. Die Promotorregionen beider Gene wurden in Abhängigkeit von bereits in der Literatur beschriebenen DNA-Polymorphismen und unter besonderer Berücksichtigung epigenetischer DNA-Modifikationen mittels bisulfitspezifischer Sequenzierung und Luciferase-Assay funktionell charakterisiert. Es konnte gezeigt werden, dass DNA-Polymorphismen und epigenetische Veränderungen der Erbinformation - letztere können in Abhängigkeit unterschiedlicher Lebenserfahrungen entstehen - funktionelle Relevanz für die Promotoraktivität der untersuchten Gene haben. Strukturelle und modifikatorische DNA-Variationen sowie Gen-Umwelt Wechselwirkungen beeinflussen somit die Genregulation und können unter bestimmten Bedingungen krankheitsrelevant werden. N2 - In this study, the 5'-regulatory regions of the genes encoding the potassiumchloride cotransporter 3 (KCC3, SLC12A6) and the glucocorticoid receptor (NR3C1) were investigated. Previously, both genes have been described to be associated with psychiatric disorder. Allelic promoter activities of the genes were investigated in dependence of DNA-polymorphisms located in the specific 5'-regulatory regions with special respect to the respective promoter methylation patterns by performing bisulfite-specific sequencing und luciferase assays. We demonstrated that genetic variation may lead to additional epigenetic modification, with the latter being susceptible to environmental factors, too. DNA-polymorphisms and epistructure were found to be of functional relevance with respect to the promoter activities of both genes. Thus, structural and epigenetic DNA variation as well as gene x environment interactions are influencing gene regulation and could lead to differential disease vulnerability under certain life conditions.
