The present thesis investigated this relationship in young healthy male subjects. Special emphasis was put on measures of HC volume and function derived from structural and functional magnetic resonance imaging (MRI). Higher cortisol levels after awakening were observed in subjects with higher levels of depressive symptomatology. Larger HC volume was associated with higher cortisol levels after awakening and in response to acute stress, whereas cognitive performance was impaired in subjects with larger HC volumes. Hippocampal activation during picture encoding was reduced after stress induction, and positive associations between activation and cognitive performance before stress were not present anymore afterwards. The present findings underscore the importance of structural and functional brain imaging for psychoneuroendocrinological research. The investigation of the association between cortisol levels and hippocampal integrity in young healthy subjects elicited unexpected results and adds to the understanding of HPA dysfunction and HC atrophy in clinical and aged populations. N2 - Eine große Anzahl von Studien beschreibt einen Zusammenhang zwischen chronischer Dysregulation der Hypothalamus-Hypophysen-Nebennieren-Achse (HHNA), Atrophy des Hippocampus (HC) sowie kognitiven- und Stimmungsveränderungen in klinischen Populationen und im Alter. Die vorliegende Arbeit untersuchte diese Zusammenhänge in jungen gesunden Männern. Einen besonderen Stellenwert im Rahmen dieser Studien nahmen Messungen von HC Volumen und -Aktivierung mit Hilfe struktureller und funktioneller Magnetresonanztomographie ein. Höhere Cortisolwerte nach dem Aufwachen standen in Zusammenhang mit depressiver Symptomatik. Höheres HC Volumen war assoziiert mit einer stärkeren Cortisolreaktion auf das Erwachen und nach akutem Streß. Die kognitive Leistung war verringert in Probanden mit höherem Hippocampusvolumen. Hippocampale Aktivierung während der Enkodierung von Bildern war reduziert nach Streßinduktion. Positive Korrelationen zwischen HC Aktivierung und Gedächtnisleistung vor der Streßaufgabe konnten im Anschluß an den Stressor nicht mehr beobachtet werden. Die vorliegenden Befunde zeigen das Potential von strukturellen und funktionellen Bildgebungsverfahren für die psychoneuroendokrinologische Forschung. Die Untersuchung des Zusammenhangs zwischen Cortisol und hippocampaler Integrität in jungen gesunden Probanden erbrachte zum Teil unerwartete Ergebnisse und wird als Beitrag zum Verständnis von Störungen der HHNA und hippocampaler Atrophy in klinischen Populationen und im Alter angesehen.
