TY - THES A1 - Vernocchi, Sara T1 - Function, detection and transcriptional origin of the membrane isoform of the Glucocorticoid Receptor T1 - Funktion, Detektion und transkriptioneller Ursprung der membranen Isoform des Glucocorticoidrezeptors N2 - Um Aufschluss über die biologische Funktion des Membran-gebundenen Glucocorticoidrezeptors (mGR) zu geben, wurden in der vorliegenden Arbeit Veränderungen im Proteom analysiert, hervorgerufen durch 15 min akuten Stress in vivo und durch kurze in vitro Aktivierung des mGR in T-Lymphozyten. Die zahlreichen Überlappungen in den zwei Datensätzen weisen darauf hin, dass der mGR in physiologisch relevanten Aktionen vermittelt und an der frühen Stressreaktion beteiligt ist, indem er schnell frühe grundlegende Ereignisse auslöst und somit den Weg für die langsameren genomischen GR Aktivitäten ebnet. Des Weiteren wird über eine neue kommerziell erhältliche Methode berichtet, welche über die entsprechende Sensitivität verfügt, um den mGR zu detektieren. Außerdem wurde der transktiptionelle Ursprung dieses Proteins untersucht. Unsere Ergebnisse deuten darauf hin, dass spezielle GR Transkripte, welche Exon 1C und 1D beinhalten, an der Expression der membranen Isoform beteiligt sind. N2 - In this thesis, in order to shed light on the biological function of the membrane-bound Glucocorticoid Receptor (mGR), proteomic changes induced by 15 min in vivo acute stress and by short in vitro activation of the mGR were analyzed in T-lymphocytes. The numerous overlaps between the two datasets suggest that the mGR mediates physiologically relevant actions and participates in the early stress response, triggering rapid early priming events that pave the way for the slower genomic GC activities. In addition, a new commercially available method with suitable sensitivity to detect the human mGR is reported and the transcriptional origin of this protein investigated. Our results indicates that specific GR-transcripts, containing exon 1C and 1D, are associated with the expression of this membrane isoform.
