Genetic analysis of Microtus arvalis from different land-use types

  • The aim of this work was to find out if all kinds of agriculture have some effects on the genetic of Microtus arvails. In this work it will be required that if genetical changes appear it will be manifested in the population. This means again that there would result different populations on different kinds of agriculture. To find out the distinction between the population two different applications were used. The multi-locus DNA fingerprinting is a method which belongs to the most evident ones. The fingerprints of this method have a high degree of information and are absolutly species specific. The disadvantage is that it- very time consuming. The other method is the RAPD-PCR. The advantage of the RAPD-PCR is caused by very fast results and easy using. The disadvantage is the less validity. Till now none of this methods have been put into practise for Microtus arvalis, so that we had to modify both ones for the field vole. Altogether 161 Microtus arvalis from Wahlen (Northern Saarland) and Herl (Trier) have been analyzed. During this it turned out that the fallow lands in Wahlen (Zill1, Zill2 and Zill3) which are in a distance between about 20m and 200m, have a homogen population regarding the field vole. In contrast to this in the fallow land at Hohberg which is about 400m away and is separated by a road, a forst and some hedges there is a field vole population which can be exactly distinguished geneticaly. However in Herl it- not possible to explain the dictinctions between the populations with distances and barriers of isolation. So that it seems to be the fact that the neighbouring fields in Herl which are not isolated by visible barriers, but just have different form of agriculture produce defferent populations of the field vole.
