Vor dem Hintergrund mehrerer Jahrhunderthochwasser im letzten Jahrzehnt werden Aspekte des präventiven (dezentralen) Hochwasserschutzes diskutiert. Dabei wird die gegenwärtige Intensität der Landnutzung mit einem verringerten Retentionsvermögen der Landschaft in Verbindung gebracht; in Extensivierungsmaßnahmen wird ein entsprechendes Potenzial zur Vermeidung von Hochwasserrisiken gesehen. Dabei ist zu beachten, dass einerseits hydrologische Simulationsmodelle zur Bewertung möglicher Landnutzungsszenarien eingesetzt werden, andererseits aber auf wissenschaftlicher Seite Defizite beim grundlegenden Erkenntnisstand über die Abflussbildung zu konstatieren sind. Der Fokus der vorliegenden Arbeit lag auf der grundlegenden Prozessforschung zur Abflussbildung in der Mikroskale und griff methodisch die Forderung nach mehr Erkenntnissen aus Geländeuntersuchungen auf. Hierzu wurden Beregnungsversuche und bodenkundliche Untersuchungen in zwei unterschiedlichen Einzugsgebieten der Trier-Bitburger Mulde durchgeführt, wodurch die substratbedingten dominanten Abflussbildungsprozesse identifiziert werden konnten. Bei dem schluffig-sandigen Substrat war die hohe texturbedingte Grobporosität für die Tiefensickerung entscheidend. Für das lehmig-tonige Substrat hingegen waren laterale Fließprozesse entlang von Regenwurmröhren über dichten Unterböden charakteristisch. Die Landnutzung führte zur Modifikation des substratspezifischen Abflussprozesses, indem sie vor allem Einfluss auf die Infiltrationseigenschaften der Bodenoberfläche nahm. Die hohe Verschlämmungsneigung des schluffig-sandigen Substrates führte zu Oberflächenabfluss unter Ackernutzung. Auf extensiv genutzten Grünlandstandorten hatte ein dichtes Geflecht von Feinwurzeln, abgestorbener Blattbiomasse und Moosen eine deutliche Reduktion der Infiltration zur Folge, so dass Oberflächenabfluss als dominanter Prozess auftrat. Auch Wälder verfügen nicht unweigerlich über ein großes Retentionsvermögen, wenn sie auf Ungunststandorten verbreitet sind. Auf dem untersuchten lehmig-tonigen Substrat wiesen die flachgründigen Waldstandorte die höchsten Abflusskoeffizienten an Subsurface Flow auf. Um die Abflussprozesse eines Landschaftsausschnittes zu charakterisieren, wurden quantifizierbare Kriterien zur flächendistributiven Ausweisung dominanter Einflussfaktoren auf die Abflussprozessbildung in einem Entscheidungsbaum zusammengefasst. Hierdurch können räumlich homogene Einheiten von hydrologischen Prozessen oder Komponenten des Wasserhaushaltes ausgewiesen werden. Die Identifikation von Abflussprozessen am Ort ihrer Entstehung ermöglicht die Planung flächenbezogener Maßnahmen des dezentralen Hochwasserschutzes
This thesis is focused on improving the knowledge on a group of threatened species, the European cave salamanders (genus Hydromantes). There are three main sections gathering studies dealing with different topics: Ecology (first part), Life traits (second part) and Monitoring methodologies (third part). First part starts with the study of the response of Hydromantes to the variation of climatic conditions, analysing 15 different localities throughout a full year (CHAPTER I; published in PEERJ in August 2015). After that, the focus moves on identify which is the operative temperature that these salamander experience, including how their body respond to variation of environmental temperature. This study was conducted using one of the most advanced tool, an infrared thermocamera, which gave the opportunity to perform detailed observation on salamanders body (CHAPTER II; published in JOURNAL OF THERMAL BIOLOGY in June 2016). In the next chapter we use the previous results to analyse the ecological niche of all eight Hydromantes species. The study mostly underlines the mismatch between macro- and microscale analysis of ecological niche, showing a weak conservatism of ecological niches within the evolution of species (CHAPTER III; unpublished manuscript). We then focus only on hybrids, which occur within the natural distribution of mainland species. Here, we analyse if the ecological niche of hybrids shows divergences from those of parental species, thus evaluating the power of hybrids adaptation (CHAPTER IV; unpublished manuscript). Considering that hybrids may represent a potential threat for parental species (in terms of genetic erosion and competition), we produced the first ecological study on an allochthonous mixed population of Hydromantes, analysing population structure, ecological requirements and diet. The interest on this particular population mostly comes by the fact that its members are coming from all three mainland Hydromantes species, and thus it may represent a potential source of new hybrids (CHAPTER V; accepted in AMPHIBIA-REPTILIA in October 2017). The focus than moves on how bioclimatic parameters affect species within their distributional range. Using as model species the microendemic H. flavus, we analyse the relationship between environmental suitability and local abundance of the species, also focusing on all intermediate dynamics which provide useful information on spatial variation of individual fitness (CHAPTER VI; submitted to SCIENTIFIC REPORTS in November 2017). The first part ends with an analysis of the interaction between Hydromantes and Batracobdella algira leeches, the only known ectoparasite for European cave salamanders. Considering that the effect of leeches on their hosts is potentially detrimental, we investigated if these ectoparasites may represent a further threat for Hydromantes (CHAPTER VII; submitted to INTERNATIONAL JOURNAL FOR PARASITOLOGY: PARASITES AND WILDLIFE in November 2017). The second part is related to the reproduction of Hydromantes. In the first study we perform analyses on the breeding behaviour of several females belonging to a single population, identifying differences and similarities occurring in cohorting females (CHAPTER VIII; published in NORTH-WESTERN JOURNAL OF ZOOLOGY in December 2015). In the second study we gather information from all Hydromantes species, analysing size and development of breeding females, and identifying a relationship between breeding time and climatic conditions (CHAPTER IX; submitted to SALAMANDRA in June 2017). In the last part of this thesis, we analyse two potential methods for monitoring Hydromantes populations. In the first study we evaluate the efficiency of the marking method involving Alpha tags (CHAPTER X; published in SALAMANDRA in October 2017). In the second study we focus on evaluating N-mixtures models as a methodology for estimating abundance in wild populations (CHAPTER XI; submitted to BIODIVERSITY & CONSERVATION in October 2017).
Energy transition strategies in Germany have led to an expansion of energy crop cultivation in landscape, with silage maize as most valuable feedstock. The changes in the traditional cropping systems, with increasing shares of maize, raised concerns about the sustainability of agricultural feedstock production regarding threats to soil health. However, spatially explicit data about silage maize cultivation are missing; thus, implications for soil cannot be estimated in a precise way. With this study, we firstly aimed to track the fields cultivated with maize based on remote sensing data. Secondly, available soil data were target-specifically processed to determine the site-specific vulnerability of the soils for erosion and compaction. The generated, spatially-explicit data served as basis for a differentiated analysis of the development of the agricultural biogas sector, associated maize cultivation and its implications for soil health. In the study area, located in a low mountain range region in Western Germany, the number and capacity of biogas producing units increased by 25 installations and 10,163 kW from 2009 to 2016. The remote sensing-based classification approach showed that the maize cultivation area was expanded by 16% from 7305 to 8447 hectares. Thus, maize cultivation accounted for about 20% of the arable land use; however, with distinct local differences. Significant shares of about 30% of the maize cultivation was done on fields that show at least high potentials for soil erosion exceeding 25 t soil ha−1 a−1. Furthermore, about 10% of the maize cultivation was done on fields that pedogenetically show an elevated risk for soil compaction. In order to reach more sustainable cultivation systems of feedstock for anaerobic digestion, changes in cultivated crops and management strategies are urgently required, particularly against first signs of climate change. The presented approach can regionally be modified in order to develop site-adapted, sustainable bioenergy cropping systems.
Die Notwendigkeit einer ökologischen Aufwertung kleiner urbaner Gewässer und damit einer effektiven Siedlungswasserwirtschaft ist in den Fokus gerückt und wurde zum Bestandteil der Wasserrahmenrichtlinie ( 2000/60/EG). Die Zunahme von Regenwassermanagementsystemen und die Renaturierung kanalisierter Gewässerabschnitte führt zu einer Integration von Flüssen und Bächen in das städtische Umfeld. Diese Arbeit fokussiert die zeitlich hochaufgelöste Analyse des Stofftransports während Abflussereignissen in mesoskaligen Einzugsgebieten. Es wird untersucht, ob die Landnutzung und die Entwässerungssysteme von Siedlungsflächen, die Abflusssdynamik und die Stoffkonzentrationen beeinflussen. Beleuchtet wird, inwieweit naturnahe Regenwassermanagementsysteme in Siedlungsgebieten den natürlichen Wasserhaushalt erhalten und eine Gewässerbelastung reduzieren. Untersucht werden die Schadstoffdynamiken und -frachten dreier verschiedener Einzugsgebiete, ähnlich hinsichtlich Einzugsgebietsgröße, Topographie und klimatischen Bedingungen, aber unterschiedlich in Landnutzung und Entwässerungssystemen. Darüber hinaus wird der Einfluss der Ableitungsart des Regenwassers aus den Siedlungsgebieten auf die Abflussdynamik und die Schadstoffverlagerung auf unterschiedlichen Skalen untersucht. Abflussganglinien und Chemographen aller Gewässer zeigen große Schwankungen während und zwischen Hochwasserereignissen. Kurzfristige Konzentrationen sind sehr hoch und können nur mittels zeitlich hochaufgelöster Beprobung bestimmt werden. Frachten und mittlere Ereigniskonzentrationen weisen eine hohe Variabilität in Abhängigkeit von vorangehenden hydro-klimatischen Bedingungen auf. Erhöhte Schadstoffkonzentrationen treten in einem ländlich geprägten Einzugsgebiet in Verbindung mit Regenfällen durch Remobilisierung von Schadstoffen aus der Kanalisation auf. In einem städtisch geprägten Vorfluter sind Schadstoffbelastungen mit dem Basisabfluss aufgrund sanitärer Fehlanschlüsse und Kläranlageneinleitungen das ganze Jahr über bedeutsam. Ein biologischer Test zur Bestimmung östrogener Aktivität zeigt, dass die Toxizität von der Kläranlagentechnologie und deren Elimination östrogener Wirkstoffe abhängt. Saisonale Effekte, durch steigende östrogene Aktivität im Sommer und abnehmende Aktivität im Winter aufgrund der Verdünnung durch Boden- und Grundwasser, konnten beobachtet werden. Dezentrale Regenrückhalte führen zum Erhalt des quasi-natürlichen Wasserhaushaltes und zur Dämpfung von Abflussspitzen im Vorfluter. Die Beprobung der Zuläufe und des Ablaufes einer Retentionsfläche zeigt die Reduktion der Maximalkonzentrationen sowie der jährlichen Frachten von Xenobiotika. Mit zunehmender Einzugsgebietsgröße nimmt die Anzahl und Diversität der Entwässerungssysteme zu. Die Skalenabhängigkeit der Gewässerbelastung ergibt sich aus dem Vorhandensein und Zusammenwirken von Misch- und Trennkanalisationen, sowie Regenwassermanagementsysteme, und Kläranlageneinläufen. Vor allem Mischwasserkanalüberläufe führen zu einer hydraulischen Belastung sowie zu hohen Momentankonzentrationen der Xenobiotika. Regenwassermanagementsysteme tragen zu einer deutlichen Reduktion der chemischen und hydraulischen Belastung des Gewässers bei. Dies führt zu einer Entlastung der Vorfluter und zu einem besseren ökologischen Zustand, wie durch die Wasserrahmenrichtlinie gefordert.
The main objective of the present thesis was to investigate whether antibody effects observed in earlier in vitro studies can translate into the protection against chemical carcinogenesis in vivo as the basis of an immunoprophylactic approach against carcinogens. As model for chemical carcinogenesis, we selected B[a]P the prototype polycyclic aromatic hydrocarbon (PAH), an environmental pollutant emanating from both natural and anthropogenic sources. Many in vivo models conveniently use high doses of carcinogens mostly given as single bolus, which provides simple surrogate readouts, but poorly reflects chronic exposure to the low concentrations found in the environment. In addition, these concentrations cannot be matched with equimolar antibody concentrations obtained by immunisation. However, low B[a]P concentrations do not permit to directly measure chemical carcinogenesis. Therefore, in the present thesis, the pharmacokinetic, metabolism and B[a]P mediated immunotoxicity were chosen as experimental read-outs. B[a]P conjugate vaccines based on ovalbumin, tetanus toxoid and diphtheria toxoid (DT) as carrier proteins were developed to actively immunise mice against B[a]P. B[a]P-DT conjugate induced the most robust immune response. The antibodies reacted not only with B[a]P but also with the proximate carcinogen 7,8-diol-B[a]P. Antibodies modulated the bioavailability of B[a]P and its metabolic activation in a dose-dependent manner by sequestration in the blood. In order to further improve the vaccination, we replaced the protein carrier by promiscuous T-helper cell epitopes to induce higher antibody titer with increased specificity for the B[a]P hapten. We hypothesised that a reduction of B cell binding sites on the carrier, compared to whole protein carrier, should favour the activation of B cells recognising the hapten instead of the carrier protein. An internal processing of the carrier and cleavage of the B[a]P-BA and subsequent presentation of the carrier peptide by MHC II molecules to T cell receptor should induce a B cell dependent immune response by activating B cells capable to recognise B[a]P. We demonstrated that a vaccination against B[a]P using promiscuous T-helper cell epitopes as a carrier is feasible and some tested peptide conjugates were more immunogenic as whole protein conjugates with increased specificity. We showed that vaccination against B[a]P reduces immunotoxicity. B[a]P suppressed the proliferative response of both T and B cells after a sub-acute administration, an effect that was completely reversed by vaccination. In immunized mice the immunotoxic effect of B[a]P on IFN-γ, Il-12, TNF-ï¡ production and B cell activation was restored. In addition, specific antibodies inhibited the induction of Cyp1a1 by B[a]P in lymphocytes and Cyp1b1 in the liver, enzymes that are known to convert the procarcinogen B[a]P to the ultimate DNA-adduct forming metabolite, a major risk factor of chemical carcinogenesis. In order to replace Freund adjuvant and to improve the immunisation strategy in terms of antibody quantity and quality, several adjuvants that are potentially compatible with their use in humans were tested. In combination with Freund adjuvant, the conjugate-vaccine induced high levels of B[a]P-specific antibodies. We showed that all adjuvants tested induced specific antibodies against B[a]P and 7,8-diol-B[a]P, its carcinogenic metabolite. The highest antibody levels were obtained with Quil A, MF-59 and Alum. Biological activity in terms of enhanced retention of B[a]P was confirmed in mice immunised with Quil A, Montanide, Alum and MF-59. Our findings demonstrate that a vaccination against B[a]P is feasible in combination with adjuvants licensed in humans. Based on these results and with the current understanding of the mechanisms of chemical carcinogenesis of the ubiquitous carcinogen B[a]P and of the effects of specific antibodies, an immunoprophylactic approach against chemical carcinogenesis is absolutely warranted. Nevertheless, the direct effects of B[a]P-specific antibodies on the different stages of carcinogenesis (e.g. adduct formation) and whether these effects may translate into long-term protective effect against tumourigenesis needs to be proven in further experiments.
Hydrodynamic processes play a fundamental role in the distribution of salt within mangrove-fringed estuaries and mangrove forests. In this thesis, two hydrodynamic processes and their ecological implications were examined. (1) Passive Irrigation and Functional Morphology of Crustacean Burrows in Rhizophora-forests. The mangrove Rhizophora excludes more than 90% of the seawater salt at water intake at the roots. By means of conductivity methods and resin casting, it was found that crustacean burrows play a key role in the removal of excess salt from the root zone. Salt diffuses from the roots into the burrows, and is efficiently flushed from the burrows by rainwater infiltration and tidal irrigation. The burrows contribute significantly to favourable conditions for the growth of Rhizophora trees. (2) Trapping of Mangrove Propagules due to Density-driven Secondary Circulation in Tropical Estuaries. In North East Australian estuaries, mangrove propagules are drifted upstream by density-driven axial surface convergences. Propagules accumulate in hydrodynamic traps upstream from suitable habitat, where they are trapped at least for the entire tropical dry season. Axial convergences may provide an efficient barrier for propagule exchange across estuaries. In such estuaries, mangrove populations can be regarded as floristically isolated, not unlike island communities, even though the populations lie on a continuous coastline. This effect may contribute to the disjunct distribution observed in some mangrove species. The outcomes of this work contribute to the understanding of the importance of salt as a growth and habitat-restricting factor in the mangrove environment.
Anpassung an den Klimawandel stellt eine komplexe gesellschaftliche Herausforderung dar und hat Bezug zu steuerungstheoretischen Fragen um Governance. Klimaanpassung zeichnet sich aus durch die Zusammenarbeit staatlicher und nicht-staatlicher Akteure, netzwerkartige Strukturen, flexible Steuerungsmechanismen sowie formelle und informelle Koordinationsstrukturen. Für die erfolgreiche Gestaltung von Klimaanpassungspolitik müssen vielfältige Akteurs- und Interessenskonstellationen berücksichtigt werden.
Ziel der vorliegenden Studie ist es, das Traben-Trarbacher Akteurs- und Stakeholdernetzwerk aus Perspektive der Klimaanpassung zu analysieren. Ein besonderer Fokus liegt hierbei auf den regionalwirtschaftlich bedeutenden Sektoren Weinbau und Tourismus, die integriert und im Kontext von kommunalen, regionalen und überregionalen Strukturen betrachtet werden. Im Rahmen der Analyse wurden das Beziehungsgeflecht, die Reichweite und Diversität des Netzwerks sowie die Zusammensetzung der Akteurslandschaft dargestellt. Darüber hinaus konnten wichtige Schlüsselakteure, potenzielle Multiplikatoren, Interdependenzen zwischen Weinbau und Tourismus sowie Informations- und Wissensquellen identifiziert werden.
Die Ergebnisse der Stakeholderanalyse geben wichtige Hinweise darauf, welche Akteure in Steuerungsprozesse von Klimaanpassung einbezogen und welche lokalen Gegebenheiten und Beziehungen hierbei berücksichtig werden müssen. Besonders die Zusammensetzung der Akteure hat entscheidenden Einfluss auf den Verlauf und Erfolg der Steuerung von Klimaanpassung. Die vorliegende Stakeholderanalyse schafft also eine wichtige Grundlage zur Etablierung eines Governance-Netzwerks für die Erarbeitung und Erprobung von Klimawandelanpassungsmaßnahmen in Traben-Trarbach und der Moselregion. Damit dient die Analyse der langfristigen Verankerung von Klimaanpassung in der Region und kann auch als Anregung für weitere Kommunen genutzt werden, die vor ähnlichen Herausforderungen stehen wie Traben-Trarbach.
Global change, i.e. climate and land use changes, severely impact natural ecosystems at different scales. Poikilothermic animals as butterflies, amphibians and reptiles have proven to be useful indicators for global change impacts as their phenology, spatial distribution, individual fitness and survival strongly depend on external environmental factors. In this aspect, phenological changes in terms of advanced flight or breeding periods, immigrations of foreign species, range shifts concomitant with temperature increases and even local population declines have been observed in both species groups. However, to date much attention has been paid to global change impacts on the species or population level and analyses concerning entire ecosystems are scarce. Applying a novel statistical modelling algorithm we assessed future changes in the extent and composition of terrestrial ecoregions as classified by the World Wide Fund for Nature (WWF). They are defined as coarse-scale conservation units containing exceptional assemblages of species and ecological dynamics. Our results demonstrate dramatic geographical changes in the extent and location of these ecoregions across all continents and even imply a repriorisation of conservation efforts to cope with future climate change impacts on biodiversity. On the local scale, climate change impacts become unequivocal. Comparing historical to contemporary butterfly assemblages on vineyard fallows of the Trier Region, a significant decline in butterfly richness, but also a severe depletion in trait diversity was observed. Comparisons of community temperature indices reveal a striking shift in community composition leading to a replacement of sedentary and monophagous habitat specialists by ubiquitous species. Similar changes have been observed in nature reserves in the Saar-Mosel-area. Monitoring data reveal strong losses of species diversity and remarkable shifts of community compositions at the expense of habitat specialists. Besides climatic variability, these findings are largely attributed to changes in habitat structures, mostly due to eutrophication and monotonisation. Management activities are unlikely to counterbalance these effects, thus severely questioning current conservation strategies. Most dramatic global change impacts are suspected on closely associated species and disruptions of biotic interactions are often hold responsible for species declines. A strong host-parasite association has developed in Myrmica ants and Maculinea butterflies, the later crucially depending on specific host ants for their larval survival. Applying environmental niche models we determined considerable niche dynamics in the observed parasite-host relation with a pronounced niche plasticity in the butterfly species adapting to previous evasive niche shifts in their host ants. Moreover, the new emergence of species continuously expanding their northernmost range borders concomitant with global warming like the Short-tailed blue (Cupido argiades) is attributed to climate change. However, species distribution models predict a severe habitat loss and shifts of potentially suitable habitats of this species towards north-eastern Europe and higher altitudes under several IPCC scenarios making the presence of this species in the Trier region a contemporary phenomenon. Species distribution models have emerged as powerful tools to predict species distributions over spatial and temporal scales. However, not only the presence of a species, but also its abundance have significant implications for species conservation. The ability to deduce spatial abundance patterns from environmental suitability might more efficiently guide field surveys or monitoring programs over large geographical areas saving time and money. Although the application of species distribution models to deduce vertebrate abundances is well recognized, our results indicate that this method is not an adequate approach to predict invertebrate abundances. Structural and ecological factors as well as climatic patterns acting at the microscale are key drivers of invertebrate occurrence and abundances limiting conclusions drawn from modeling approaches. Population declines should be interpreted with care as in butterflies and amphibians various reasons are debated. Both species groups are acknowledged to be highly susceptible to land use changes and variations in landscape structure. Moreover, climate and land use are not independently operating factors. The combined impact of both is demonstrated in our study linking climate-driven changes in amphibian phenologies to temporal advanced applications of pesticides and fertilizers. Both environmental factors already represent severe threats to amphibians when standing alone, but linking their combined impacts may result in an potentiated risk for amphibian populations. As all amphibians and numerous butterfly species are legally protected under the Federal Nature Conservation Act, intensifications of agricultural land use in large parts of Germany as well as new agrarian practices (including genetically manipulated plants accompanied by new herbicide technologies) might severely challenge regional conservation activities in the future.
Geographic ranges of species and their determinants are of great interest in the field of biogeography and are often studied in terms of the species" ecological niches. In this context, the range of a species is defined by the accessibility of an area, abiotic factors and biotic interactions, which affect a species" distributions with different intensities across spatial scales. Parapatry describes a distributional pattern in which the ranges of two species meet along sharp range limits with narrow contact zones. Such parapatric range limits are determined by changing abiotic conditions along sharp environmental gradients or can result from interspecific resource competition. However, it has been shown that often the interplay of abiotic conditions and species interactions determine parapatry. The geographic ranges of the land salamanders, Salamandra salamandra and S. atra, narrowly overlap in the European Alps with only few syntopic localities and to date, the cause of parapatry is unknown. The goal of this thesis was thus to identify the importance of abiotic and biotic factors for their parapatric range limits at different spatial scales. On a broad spatial scale, the role of climate for the parapatric range limits of the species was investigated within three contact zones in Switzerland. Climatic conditions at species" records were analysed and species distribution modelling techniques were used to explore the species" climatic niches and to quantify the interspecific niche overlap. Furthermore, it was tested whether the parapatric range limit coincides with a strong climatic gradient. The results revealed distinct niches for the species as well as the presence of strong climatic gradients which could explain the parapatric range limits of the species. Yet, there was a moderate interspecific niche overlap in all contact zones indicating that the species may co-occur and interact with each other in areas where they both find adequate conditions. Comparison among contact zones revealed geographic variation in the species" niches as well as in the climatic conditions at their records suggesting that the species can occur in a much wider range of conditions than they actually do. These findings imply that climate represents a main factor for the species" parapatric range limits. Yet, interspecific niche overlap and the geographic variation provide indirect evidence that interspecific interaction may also affect their spatial distribution. To test whether competition restricts the species" ranges on the habitat scale and to understand local syntopic co-occurrence of the salamanders within their contact zones, site-occupancy modelling was used. This approach allowed to find the habitat predictors that best explain the species" local distribution. While the slope of the site positively affected the occupancy probability of S. salamandra, no tested predictor explained that of S. atra. Also, there was no effect of the occurrence of one species on the occupancy probability of the other providing no evidence for competition. Should competition occur, it does not lead to spatial segregation of the species on this scale. Because biotic interactions most significantly affect the ranges of species on small spatial scales, the microhabitat conditions at locations of the species within syntopic contact zones were compared and a null model analysis was applied to determine their niche overlap. Resource selection probability function models were used to assess those attributes that affect the species" habitat selections. The results revealed species-specific microhabitat preferences related to leaf litter cover, tree number and that the species were active at different temperatures as well as times of the day. The high degree of diurnal activity of S. atra may be due to its preference of forest floor microhabitats that long remain suitable during daytime. Besides, there was a great niche overlap for shelters indicating that the species may compete for this resource. Differential habitat selection and the use of the available shelters at different times of the day may minimize species interactions and allow their local co-occurrence within contact zones. To identify whether the potential infection with the pathogenic chytrid fungus could serve as an alternative biotic explanation for the range margins of S. atra, several populations throughout its range were screened for infection. Since the occurrence of this pathogen was detected mostly at lower altitudes of the Alps, it may confine the range of S. atra to higher elevations. Because chytrid was not detected in any of the samples, the pathogen unlikely plays a role in determining its range limits. Overall, these findings underline the complexity of mechanisms that determine the range margins of parapatric species and provide an important basis for subsequent studies regarding the determinants of the parapatric distribution of the two salamander species.
Die Region Trier ist ein seit langem intensiv besiedelter Kulturraum. In der vorliegenden Arbeit galt es, unterschiedliche morphodynamische Prozessphasen während des Holozäns zu unterscheiden und ihre Ursachen zu klären. Gegenstand der Untersuchungen waren Schwemmfächer und andere Sedimentkörper kleiner Einzugsgebiete, die als korrelate Sedimentkörper der (Boden-)Erosion in den Einzugsgebieten betrachtet werden. Mit Hilfe sedimentologisch-geochemischer Untersuchungen und Datierungen der Sedimente wurde der Zusammenhang zwischen dem Erosions- und Akkumulationsgeschehen, der Landnutzungsintensität und der klimatischen Entwicklung während des Holozäns untersucht. Der menschliche Einfluss in den Einzugsgebieten wurde über archäologische Funde des Rheinischen Landesmuseums Trier und historische Karten abgeglichen. Der klimatische Einfluss wurde mangels ausreichender regionaler Datendichte über die bekannte überregionale Klimaentwicklung des Holozäns in Mitteleuropa abgeschätzt. Es konnten Aktivitätsphasen (vorgeschichtlich, (früh-)römisch, Mittelalter) und Stabilitätsphasen (Völkerwanderungszeit) ausgegliedert werden. Die unterschiedlichen morphodynamischen Prozessphasen haben ihren Ursprung in der lokalen anthropogenen Landnutzung in Kombination mit dem Witterungs- und Wettergeschehen. Die Bedeutung des Klimas mit seinen gemittelten Werten holozäner "Optima" und "Pessima" tritt dahinter zurück. Die Schwermetallgehalte von Nickel, Kupfer, Zink und Blei in den fossilen und rezenten Böden der Region sind stärker von lokalen als von globalen Faktoren beeinflusst, wie sie an Geoarchiven ohne eigenen geogenen background nachgewiesen sind. Der Einsatz geochemischer Indizes ermöglicht in seltenen Fällen eine weitere Untergliederung makroskopisch homogener Sedimente. Ihre inhaltliche Interpretation wird durch die Tatsache eingeschränkt, dass es sich bei Schwemmfächern um offene Systeme handelt, bei denen in humiden Klimaten nicht entschieden werden kann, welche Verwitterungsprozesse prä- und welche postsedimentärer sind.
Soil and water conservation are cross-sectional assignments. The respective objectives of the individual interest groups cause conflicts of use and lead to different assessments of the soil's potential. Necessary decisions and the practical implementation of soil and water conservation measures require the use of data. These data, which are both spatial and temporal, characterise past, present and, in the case of predictions, also future environmental conditions. The multitude of relevant data necessitates the use of geographic information systems as an instrument for successful resource management. With the use of problem-oriented case studies, it was possible to show that an improved understanding of the system is necessary for both optimisation of the site-specific resource management within the framework of Precision Farming and for the assessment of local to regional conflicts of use with regard to land usage and soil and water conservation. By changing the method, sufficient respective measures regarding documentation, prevention and risk assessment were able to be introduced and implemented. With the objective of practical implementation of a sustainable resource management, the possibilities of short- to long-term initiation of self-organised systems through the networking of available (geo-)information as well as the respective interest groups involved in the conflict of use formed the focal point of this investigation. The creation of networks linking agriculture, water extractors and nature conservation promotes necessary synergies and emergences, due to increased communication. Not the conveyance of knowledge alone, but rather new forms of understanding cause the interest groups involved to change their behaviour, thus facilitating efficient resource management for the interests of soil and water conservation.
Ziel der Optimierung bestimmter molekulargenetischer Methoden war die Aufklärung möglicher Zusammenhänge zwischen landwirtschaftlichen Nutzungsformen und der Genetik von Microtus arvalis. Um mögliche Populationsunterschiede feststellen zu können wurden zwei unterschiedliche genetische Untersuchungsmethoden eingesetzt. die Fingerprints, die mit dem Multi-locus DNA Fingerprint gemacht werden besitzen einen sehr hohen Informationswert und sind individualspezifisch. Der Nachteil dieser Methode besteht darin, dass sie sehr aufwendig ist. Die RAPD-PCR hat den Vorteil, dass sie sehr schnell Bandemuster produziert, und leicht anwendbar ist. der Informationsgehalt des RAPD-PCR ist jedoch geringer. Da beide Methoden auf die Feldmaus noch nicht angewandt wurden, mussten sie so modifiziert werden, dass genetische Untersuchungen mit Microtus arvalis erst möglich wurden. Insgesamt wurden 161 Microtus arvalis aus Wahlen (Nordsaarland) und Herl (Regierungsbezirk trier) untersucht. Dabei stellte sich heraus, dass die Brachflächen in Wahlen (Zill1, Zill2 und Zill3), die zwischen ca. 20m und 200m voneinander entfernt liegen erwartungsgemäß eine zusammenhängende Population darstellen. Die Feldmäuse der Brachfläche am Hohberg (ca. 400m entfernt) die durch eine Straße, sowie Wald bzw. Feldgehölze von den Zill-Populationen getrennt sind können genetisch eindeutig abgegrenzt werden. Noch deutlicher sind die Populationsunterschiede von Wahlen zu Herl. Die Populationsunterschiede in Herl lassen sich weder durch die Distanz noch durch Isolationsbarrieren erklären. Alles deutet darauf hin, dass einzelne Bandenmuster mit den spezifischen Landbewirtschaftungsformen korrelieren.
Earth observation (EO) is a prerequisite for sustainable land use management, and the open-data Landsat mission is at the forefront of this development. However, increasing data volumes have led to a "digital-divide", and consequently, it is key to develop methods that account for the most data-intensive processing steps, then used for the generation and provision of analysis-ready, standardized, higher-level (Level 2 and Level 3) baseline products for enhanced uptake in environmental monitoring systems. Accordingly, the overarching research task of this dissertation was to develop such a framework with a special emphasis on the yet under-researched drylands of Southern Africa. A fully automatic and memory-resident radiometric preprocessing streamline (Level 2) was implemented. The method was applied to the complete Angolan, Zambian, Zimbabwean, Botswanan, and Namibian Landsat record, amounting 58,731 images with a total data volume of nearly 15 TB. Cloud/shadow detection capabilities were improved for drylands. An integrated correction of atmospheric, topographic and bidirectional effects was implemented, based on radiative theory with corrections for multiple scatterings, and adjacency effects, as well as including a multilayered toolset for estimating aerosol optical depth over persistent dark targets or by falling back on a spatio-temporal climatology. Topographic and bidirectional effects were reduced with a semi-empirical C-correction and a global set of correction parameters, respectively. Gridding and reprojection were already included to facilitate easy and efficient further processing. The selection of phenologically similar observations is a key monitoring requirement for multi-temporal analyses, and hence, the generation of Level 3 products that realize phenological normalization on the pixel-level was pursued. As a prerequisite, coarse resolution Land Surface Phenology (LSP) was derived in a first step, then spatially refined by fusing it with a small number of Level 2 images. For this purpose, a novel data fusion technique was developed, wherein a focal filter based approach employs multi-scale and source prediction proxies. Phenologically normalized composites (Level 3) were generated by coupling the target day (i.e. the main compositing criterion) to the input LSP. The approach was demonstrated by generating peak, end and minimum of season composites, and by comparing these with static composites (fixed target day). It was shown that the phenological normalization accounts for terrain- and land cover class-induced LSP differences, and the use of Level 2 inputs enables a wide range of monitoring options, among them the detection of within state processes like forest degradation. In summary, the developed preprocessing framework is capable of generating several analysis-ready baseline EO satellite products. These datasets can be used for regional case studies, but may also be directly integrated into more operational monitoring systems " e.g. in support of the Reducing Emissions from Deforestation and Forest Degradation (REDD) incentive. The allergic contact dermatitis (ACD) to small molecular weight compounds is a common inflammatory skin reaction. ACD is restricted to industrialized countries, has an enormous sociomedical and socioeconomic impact. About 2,800 compounds from the six million chemicals known in our environment are believed to have allergic, and to a lesser degree also contact-sensitizing or immunogenic properties causing allergic contact dermatitis. ACD results from T cell responses to harmless, low molecular weight chemicals (haptens) applied to the skin. Haptens are not directly recognized by the cells of the immune system. They need to be presented by subsets of antigen presenting cells to the cells of the immune system. In this regard, epidermal Langerhans cells (LC) and the cells into which they mature (dendritic cells) are believed to play a pivotal role in the sensitization process for ACD. LC are able to bind the haptens, internalize them, and present them to naive T cells and induce thereby the development of effector T cells. They are so-called professional antigen presenting cells. This process is initiated and maintained by the release of several mediators, which are released by various cells after their contact with the haptens. One of the first proteins secreted into the environment is interleukin (IL)-1ß. This cytokine is produced and secreted minutes after an antigen enters the cell. It is commonly believed that the large amounts of this protein and other cytokines such as granulocyte-colony stimulation factor (GM-CSF) and tumor necrosis factor alpha (TNF-ï¡) needed for the initiation and activation of ACD are coming first from other cells residing in the skin, e.g., keratinocytes, monocytes and macrophages. These cytokines provide the danger signals needed for the activation of the Langerhans cell (LC), which then produce via a positive feedback loop various cytokines themselves. In addition, other proteins such as chemokines influence the generation of danger signals, migration, homing of T cells in the local lymph nodes as well as the recruitment of T cells into the skin. Thus, a small molecular compounds or hapten needs to be able to induce danger signals in order to become immunogenic. In this study, we investigated whether para-phenylenediamine (PPD), an arylamine and common contact allergen, is able to induce danger signals and likely provide the signals needed for an initiation of an immune response[162, 163]. PPD is used as an antioxidant, an ingredient of hair dyes, intermediate of dyestuff, and PPD is found in chemicals used for photographic processing. But up to date, it has not been clearly demonstrated if PPD itself is a sensitizing agent. Thus, this study aimed on the potential of PPD to provide the danger signals by studying IL-1β, TNF-ï¡, and monocyte chemoattractant proteins (MCP-1) in human monocytes, peripheral blood mononuclear cells (PBMC) from healthy volunteers, and also in two human monocyte cell lines namely U937, and THP-1. This study found that PPD decreased dose- and time-dependently the expression and release of three relevant mediators involved in the generation of danger signals. Namely, PPD reduced the mRNA and protein levels for IL-1ß, TNF-ï¡, and MCP-1 in primary human monocytes from various donors. These findings were extended and validated by investigations using the cell line U937. The data were highly specific for PPD, and no such results were gained for its known auto oxidation product called Bandrowski- base or for meta-phenylenediamine (MPD), and ortho-phenylenediamine (OPD). Therefore, we can speculate that this effect is likely to be dependent on the para-substitution. Based on these results we conclude that PPD itself is not able to mount a cascade for the induction of danger signals. It should be mentioned that it is still possible that PPD induces danger signals for sensitization by other unknown processes. Therefore, more research is still needed focusing on this subject especially in professional antigen presenting cells in order to solve the still open question whether PPD itself sensitizes naive T cells or if PPD is solely an allergen. Independently we found unexpectedly that PPD as well as other haptens such as 2, 4-Dinitrochlorobenzene, nickelsulfate, as well as some terpenoide increased clearly the expression of CC chemokin receptor 2 (CCR2), the receptor for the chemokine MCP-1. Up to date, the main importance for the CCR2 receptor comes from results demonstrating that CCR2 is critical for the migration of monocytes after encounter with bacterial lipopolysaccharides. Under these circumstances the receptor disappears from the cell surface and is down regulated. An up regulation of CCR2 has not been reported for haptens, and deserves further investigations.
The microbial enzyme alkaline phosphatase contributes to the removal of organic phosphorus compounds from wastewaters. To cope with regulatory threshold values for permitted maximum phosphor concentrations in treated wastewaters, a high activity of this enzyme in the biological treatment stage, e.g., the activated sludge process, is required. To investigate the reaction dynamics of this enzyme, to analyze substrate selectivities, and to identify potential inhibitors, the determination of enzyme kinetics is necessary. A method based on the application of the synthetic fluorogenic substrate 4-methylumbelliferyl phosphate is proven for soils, but not for activated sludges. Here, we adapt this procedure to the latter. The adapted method offers the additional benefit to determine inhibition kinetics. In contrast to conventional photometric assays, no particle removal, e.g., of sludge pellets, is required enabling the analysis of the whole sludge suspension as well as of specific sludge fractions. The high sensitivity of fluorescence detection allows the selection of a wide substrate concentration range for sound modeling of kinetic functions.

The availability of data on the feeding habits of species of conservation value may be of great importance to develop analyses for both scientific and management purposes. Stomach flushing is a harmless technique that allowed us to collect extensive data on the feeding habits of six Hydromantes species. Here, we present two datasets originating from a three-year study performed in multiple seasons (spring and autumn) on 19 different populations of cave salamanders. The first dataset contains data of the stomach content of 1,250 salamanders, where 6,010 items were recognized; the second one reports the size of the intact prey items found in the stomachs. These datasets integrate considerably data already available on the diet of the European plethodontid salamanders, being also of potential use for large scale meta-analyses on amphibian diet.
With the ongoing trend towards deep learning in the remote sensing community, classical pixel based algorithms are often outperformed by convolution based image segmentation algorithms. This performance was mostly validated spatially, by splitting training and validation pixels for a given year. Though generalizing models temporally is potentially more difficult, it has been a recent trend to transfer models from one year to another, and therefore to validate temporally. The study argues that it is always important to check both, in order to generate models that are useful beyond the scope of the training data. It shows that convolutional neural networks have potential to generalize better than pixel based models, since they do not rely on phenological development alone, but can also consider object geometry and texture. The UNET classifier was able to achieve the highest F1 scores, averaging 0.61 in temporal validation samples, and 0.77 in spatial validation samples. The theoretical potential for overfitting geometry and just memorizing the shape of fields that are maize has been shown to be insignificant in practical applications. In conclusion, kernel based convolutions can offer a large contribution in making agricultural classification models more transferable, both to other regions and to other years.
Es wurden Staubmessungen in der Luft eines Putenmaststalls mit dem Ziel der Charakterisierung der Höhe und des Verlaufs besonders der Feinstaubfraktion PM 10 (Particulate Matter 10, Partikel, deren aerodynamischer Durchmesser kleiner als 10 -µm ist) und der einatembaren Staubfraktion (ID bzw. PM 100) im Laufe eines Mastdurchganges durchgeführt. Zur Erfassung von PM 10 wurde das für Emissionsmessungen bereits etablierte Messsystem DIGITEL HVS DHA-80 eingesetzt, um es auf Eignung unter Stallbedingungen zu testen. Die Ansaughöhe zur Probenahme betrug 2 m über Grund. Parallel dazu wurde die einatembare Staubfraktion mit einem in der Arbeitsmedizin gebräuchlichen, stationär positionierten Personal Sampler (IOM-SKC-Messsystem) erfasst. Anlass der Erfassung der Staubkonzentrationen war der unbefriedigende Kenntnisstand bezüglich der Zuordnung der Emissionen zur Quelle "Tierstall" und bezüglich der Quantitäten der Staubfrachten. Ein wesentlicher Grund für die bislang geringe Anzahl von Feinstaubmessungen in Tierhaltungen liegt in dem Fehlen standardisierter Messverfahren für diese Komponente, was zudem den Vergleich der bisherigen Messergebnisse erschwert. Der Wert der genauen Kenntnis der Konzentrationen der partikelförmigen Emissionen besonders Feinstaub (PM 10) in und aus landwirtschaftlicher Nutztierhaltung liegt in der Einschätzung des Gefährdungspotentials für den Menschen unter Berücksichtigung umwelthygienischer und umweltrechtlicher Aspekte. Letztere werden unter anderem durch das Vorsorgeprinzip im Bundes-Immissionsschutzgesetz dargestellt, wobei die Minderung der von Tierhaltungen ausgehenden Emissionen bereits wichtiger Bestandteil nationaler und europäischer Luftreinhaltepolitik ist. So beschäftigen sich Umwelt- und Arbeitsmediziner aufgrund des Verdachts erhöhter Prävalenz umweltbezogener Beschwerdebilder wie Störungen im Bereich der Atemwege in Regionen mit spezialisierter Tierhaltung und bezüglich des Auftretens als arbeitsschutzrelevanter Arbeitsstoff verstärkt mit diesem Feinstaub. Darüber hinaus werden die Daten von den Behörden als Entscheidungshilfe für die Bewilligung neuer Stallbauten benötigt, um Anwohner vor möglichen negativen Einflüssen zu bewahren. Derzeit wird davon ausgegangen, dass PM 10 aus Nutztierhaltungen, aufgrund der durch die Abstandsregelung der TA Luft (2002) greifenden Verdünnungseffekte keine negativen Auswirkungen auf die Gesundheit von Anwohnern hat. Diese Annahme ist allerdings bisher nicht durch ausreichende Messungen abgesichert. Die im Rahmen der Arbeit durchgeführten Messkampagnen haben zusammen mit der Literaturrecherche ergeben, dass das Messsystem DIGITEL HVS DAH-80 zur Erfassung von PM 10-Konzentrationen in Geflügelstallungen als geeignet bewertet werden kann. Die über 24 Stunden maximal erfasste mittlere PM 10-Konzentration beträgt 2,44 mg•m-3, der Durchschnitt liegt bei 1,27 mg•m-3, wobei die PM 10-Konzentrationen im Tagesverlauf tagesrhythmische Ereignisse widerspiegeln. Die ebenso über 24 Stunden erfassten ID-Konzentrationen liegen in Bereichen von 0,83 mg•m-3 bis 4,34 mg•m-3. Beide Staubfraktionen wurden im Hinblick auf bekannte Arbeitsplatz-Grenzwerte (AGW) sowie in Bezug auf umweltrechtliche Aspekte bewertet. Da bisher keine berufsbezogenen Grenzwerte existieren, wurden die beiden Staubkonzentrationen als arbeitsschutzrelevante Arbeitsstoffe mit den allgemein gültigen Grenzwerten der GefStoffV von E-Staub (einatembarer Staub) und A-Staub (Alveolengängiger Staub) mit einem AGW von 10 mg•m-3 bzw. 3 mg•m-3 verglichen. Selbst die bestimmten maximalen Tages-Konzentrationen liegen unterhalb dieser AGWs. Von ausgewählten Staubproben beider Fraktionen wurden die Aktivitäten der ebenso zu den umwelthygienisch bedeutenden Luftinhaltsstoffen zählenden Endotoxine ermittelt und mit bekannten Werten aus der Literatur verglichen. Für diese biologische Komponente besteht derzeit kein Grenzwert, sondern nur Orientierungswerte, die von 50 EU•m-3 bis 1.000 EU•m-3 reichen. Mehr als die Hälfte der ermittelten Endotoxinaktivitäten im E-Staub lagen unterhalb von 1.000 EU•m-3, die der analysierten PM 10-Teilfilter lagen im Bereich von 150 EU•m-3 bis 3.500 EU•m-3. Bei umwelthygienischen oder arbeitsmedizinischen Fragestellungen sollte die Qualität der Staubfraktionen zunehmend im Fokus der Messungen stehen, da diese aufgrund zahlreicher Einflussfaktoren eine erhebliche Variabilität aufweisen kann. Die weitere Bearbeitung des relativ neuen Arbeitsgebietes PM 10 im Bereich der landwirtschaftlichen Nutztierhaltung erfordert zudem eine verstärkte interdisziplinäre Zusammenarbeit und den Austausch von Erkenntnissen und Erfahrungen zwischen den beteiligten Disziplinen der Wissenschaft und Forschung aber auch mit Gesetzgebern und Genehmigungsbehörden.
The increase in recreational leisure during the last century and the increase in the individual mobility since the sixties have induced growing travel activities as well as a growing demand for recreation in a natural environment. Politically the demand for recreational leisure is regarded as a fundamental part of human life and is legally embedded as inherent goal of nature resorts besides nature conservation. Following new pedagogic principles man should get familiar with the environment in order protect nature in the long-run. Taking the bicycle concept of the nature resort Saar-Hunsrück with special emphasis on the St. Wendel country as an example the bicycle tourism which fulfils the principles of sustainability is analysed with regard to the acceptance by the tourists. Scenic and historical attractions of the area are deliberately taken into account in the design of the bicycle routes. By analysing the results of a survey it is the objective to work out applicable results for the design of routes and systems of orientation. Moreover it is the objective of a special survey on mountain bikers to answer the question on how this extreme type of biking could be incorporated in the supply of recreational activities of the nature resort and whether it can contribute to a growing environmental sensitivity.
Extension of an Open GEOBIA Framework for Spatially Explicit Forest Stratification with Sentinel-2

Spatially explicit information about forest cover is fundamental for operational forest management and forest monitoring. Although open-satellite-based earth observation data in a spatially high resolution (i.e., Sentinel-2, ≤10 m) can cover some information needs, spatially very high-resolution imagery (i.e., aerial imagery, ≤2 m) is needed to generate maps at a scale suitable for regional and local applications. In this study, we present the development, implementation, and evaluation of a Geographic Object-Based Image Analysis (GEOBIA) framework to stratify forests (needleleaved, broadleaved, non-forest) in Luxembourg. The framework is exclusively based on open data and free and open-source geospatial software. Although aerial imagery is used to derive image objects with a 0.05 ha minimum size, Sentinel-2 scenes of 2020 are the basis for random forest classifications in different single-date and multi-temporal feature setups. These setups are compared with each other and used to evaluate the framework against classifications based on features derived from aerial imagery. The highest overall accuracies (89.3%) have been achieved with classification on a Sentinel-2-based vegetation index time series (n = 8). Similar accuracies have been achieved with classification based on two (88.9%) or three (89.1%) Sentinel-2 scenes in the greening phase of broadleaved forests. A classification based on color infrared aerial imagery and derived texture measures only achieved an accuracy of 74.5%. The integration of the texture measures into the Sentinel-2-based classification did not improve its accuracy. Our results indicate that high resolution image objects can successfully be stratified based on lower spatial resolution Sentinel-2 single-date and multi-temporal features, and that those setups outperform classifications based on aerial imagery only. The conceptual framework of spatially high-resolution image objects enriched with features from lower resolution imagery facilitates the delivery of frequent and reliable updates due to higher spectral and temporal resolution. The framework additionally holds the potential to derive additional information layers (i.e., forest disturbance) as derivatives of the features attached to the image objects, thus providing up-to-date information on the state of observed forests.
